24x48 pole barn, need to pour slab first. 6x6's on top of slab?

If you have the "kit" then the 6x6 lengths or instructions will tell you the designers best way. If you do not have the 6x6 yet then think of using pipe with rebar stubs set in holes with the slab poured around them. My 30x40x12 clear span carport is on 2" schedule 40 pipe set 3' in the ground and has withstood everything up to hurricane winds. As far as the anchors forget box stores and check with metal building suppliers.

There is another way to do this.

Make the concrete driveway strong enough to support a concrete truck. There is no mystery about how to do this, the highway department in your state does it all the time.

Yup, it will cost a little more, but you will be very happy if you ever need to get something big back to your barn.

Most new construction does the driveway last. I can't think of ever seeing a driveway get poured before the house was done, or almost done.

I agree with the above. I'd not want to put a delicate driveway between my access route and my construction site. Either build a driveway suitable for the construction traffic, or build it as part of the finishing touches. Your plans are akin to getting the cart before the horse.
