I really don't want to start a big debate here on which engine or transmission is best so I'll try to answer you with what I would get for a particular purpose. Understand I'm not saying it's best, only it's what I would buy for a particular purpose based on my knowledge, experiences and biases at this moment and are subject to change as my experience changes.
For pulling loads I might otherwise try to pull with a class 3, 4, 5 or 6 truck, I'd probably go with something like a single axle day cab with no more than a nine or ten speed transmission. The super-10 would be a good way to go as would something like an Allison automatic if you could find one.
I'm in Indiana so hills aren't much of a concern at all. I can't imagine needing any more than 300 HP to do anything I'd want to do. If I had no idea what'd work for me, I'd load up my trailer and pull it around with some different HP options I was considering. Once I found that point below which I was unhappy with performance pulling my load, I'd simply avoid those when I was shopping. I don't worry about rear end gearing unless it's way out to either end of the spectrum. Something at or near 3.71 would be fine for me. As to suspension, air ride. I'd make sure I had an air ride seat, too. If I were going to have a passenger, I'd make sure I got air ride on both sides. That's pretty common, anymore, but I'd look just to make sure.
