Vapor Barrier should be applied to the 'warm side' of the heated space.
Vapor barrier will not work as intended if it is punctured.
I would recommend a closed cell poly Iso spray-in foam in between pressure treated sleepers. The closed cell foam will self seal around the sleepers and act as a vapor barrier.
Vapor Barriers are really not barriers - they are only retarders - slowing down the vapor.
The more attention to detail, the better.
