I have a John Deere 3320 with cab and R-1 tires fluid filled. My land is a bit hilly with black dirt over clay, In Iowa. Will it handle a 6" box blade?

Depends on how heavy the box is, how tough the substrate is and how your traction is. I have roughly 850# 6 footer on a 3520 and I tend to lose traction before I fill the box. Its a nice size to cover the tracks, and you should have better traction than I do. For that frame a 66" or 72" seems just right.

My 32hp 3,500lbs (loaded with losder) Kubota L3200 with R4s pulls my 72" King Kutter box. Box could be a bit heavier & a 66" probably would be a better fit, but it pulls it. Mostly road maintnance, but have done a decent bit of excavating with it. You have a beefier tractor than I do...

This is about the third or fourth thread the OP has started on box blades, so you are forgiven!
The real answer is to measure the outside width of the rear wheels, since the 3320 came several different ways -- you didn't say what configuration your R1 tires are in. The box needs to be at least that wide. If I remember right, you'd need anywhere from a 60" to a 66" minimum. I am sure it will pull a 72", but I know someone else with a 3720 that has limitations with a 72" box, so consider it an upper limit for sure.
